Repeat administration of AMG 416 results in extended reduction of PTH in renally compromised rats. Rats subjected to 5/6 nephrectomy were treated daily for 28 days with either: AMG 416 (1 mg/kg administered by IV bolus), saline (IV bolus) or cinacalcet (10 mg/kg administered po). Blood chemistries are reported relative to the last (day 28) dose. (A) Plasma PTH measured at 6, 16 and 48 hr from the last dose. (B) Plasma PTH measured at 16 hr following dosing on Days 7, 14, 21 and 28. (C) Serum calcium at pre-dose, 6 and 16 h from the last dose.
