Investigation of the recycling process of AQP2 by inhibition of endocytosis via GTPase-deficient dynamin-2 mutation (K44A) in transiently transfected, unstimulated MDCK-AQP2 cells. The MDCK cells stably expressing AQP2-WT or AQP2-R254W were transiently transfected with plasmid DNA encoding GFP-tagged dynamin-2, either Dyn2-WT (a,c) or Dyn2-K44A (b,d). Two days post-transfection the cells were immunostained with AQP2-sensitive antibody and subjected to confocal laser scanning microscopy. Note the AQP2 staining of the plasma membrane in cells expressing AQP2-WT indicated with an arrow (b). The presented data represents the results obtained in two independent experiments.
